Bioaktive anorganische Gerüststrukturen


Inorganic ScaffoldDie Entwicklung keramischer Trägerstrukturen für den Knochenersatz erfolgt in der FMZ aus reaktiven Mischungen von Calcium- und Magnesiumphosphaten, die nach Zusatz einer wässrigen Phase einer Zementreaktion unterliegen und so ohne Sinterung ein strukturell stabiles Implantat bilden.

Vorrangiges Ziel ist die Untersuchung von pulvertechnologischen Eigenschaften im Hinblick auf die Reaktivität, der Wechselwirkung der Partikel in flüssiger Phase mit Additiven, etwa zur Einstellung injizierbarer Eigenschaften sowie der mechanischen Eigenschaften der abgebundenen Zementmatrices.

Inorganic ScaffoldEinen Schwerpunkt bildet die Verwendung von Calciumphosphat-Zementen (CaP) für den 3D-Pulverdruck, was die Darstellung individueller Implantat Geometrien mit hoher Auflösung (Abbildung rechts oben) und einer gerichteten Makroporosität für die Einsprossung von Blutgefäßen ermöglicht (Abbildung links mitte).

Neuere Arbeiten beschäftigen sich mit dem Einsatz mikroporöser CaP-Werkstoffe als Träger für bioaktive Substanzen. Durch Verwendung eines Mehrkomponenten-Druckers kann gleichzeitig ortsaufgelöst die Modifikation von Strukturen mit diesen Wirkstoffen erfolgen, um sowohl eine ortsgerichtete biologische Reaktion als auch eine Steuerung der Freisetzungskinetik zu erreichen.

Eine Hauptstrategie der FMZ zur Verbesserung des typischerweise spröden Bruchverhaltens von Phosphat-Zementen besteht in der Verstärkung mit Fasern (Abbildung rechts unten). Dazu werden sowohl die Kompatibilität als auch das Abbauverhalten von Faser und Matrix optimiert und aufeinander abgestimmt.

Inorganic ScaffoldOffene Porosität ist eine Grundvoraussetzung für Vaskularisation und das Einwachsen von Zellen. Ein Ansatz der FMZ, um diese mit möglichst hoher mechanischer Stabilität zu erreichen, besteht in der Abformung von Schäumen mit einer geeigneten Nachbehandlung der Gerüste.

Elektrochemisch gestützte Abscheidung keramischer Schichten


Die Biokompatibilität vor allem nicht-abbaubarer Werkstoffe wird von ihren Oberflächeneigenschaften bestimmt. Die Methode der elektrochemischen Abscheidung (unteres Bild) wird genutzt, um metallische Oberflächen mit geringkristallinen Calcium- und Magnesiumphosphat-Schichten zu versehen. Ein Ziel dieser Arbeiten ist die Entwicklung neuartiger Mehrphasen-beschichtungen mit eingelagerten Ionen, die bakterizide und biokompatible Eigenschaften kombinieren und damit sowohl das Entzündungsrisiko nach der Operation senken als auch das Einwachsen des Implantats unterstützen können.
